Graphical abstract

During eukaryotic transcription, nascent mRNAs hybrid with the DNA template to form an R-loop structure. The THO-Sub2 complex is recruited and associates with the transcription machinery to unwind this harmful R-loop structure and ensure efficient mRNA export.
